Single Dose Oral Typhoid Vaccine Shown in US Multi-Centre Trial to be Safe and Immunogenic
UK, February 27th, 2003 โ" Microscience has announced the results of a multi-centre clinical trial which shows that their new generation single dose oral typhoid vaccine is safe, well-tolerated and highly immunogenic. The study was carried out using a freeze-dried product suitable for commercialisation. These results confirm the finding of an earlier trial in the UK.
The multi-centre outpatient trial involved 60 healthy adult volunteers and was carried out under a Microscience sponsored IND at the University of Vermont with Fletcher Allen Healthcare, Burlington VT and the Johns Hopkins University Bloomberg School of Public Health, Baltimore MD.
โThese latest most promising results move our oral typhoid vaccine centre stageโ said Research and Development Director Dr Steven Chatfield today โgiven that the freeze dried product used for this randomised, double-blind, dose escalation placebo controlled trial is that proposed for the final commercial vaccine. The fact that we already have a manufacturing process in place suitable for commercialising the product is of major strategic importance. This will result in savings, both time and money as we move this vaccine quickly through the Phase II programmeโ
Microscience are developing this live attenuated vaccine principally to provide travellers with an effective single dose vaccine, which can be taken by mouth to protect against typhoid fever. It will be a convenient new alternative to current multi-dose oral and single dose injected vaccines.
A significant number of cases of typhoid are reported in both US and Europe most of which are contracted while abroad with an estimated global incidence of 30 million cases and 600,000 mortalities, worldwide every year.
In the trial the vaccine was found to have an excellent safety profile at all three dose levels. These were 5 x 107 and 5 x 108 and 5 x 109 live attenuated bacteria. It was well tolerated with no bacteraemias attributable to the vaccine at any dose level. The incidence of adverse events was not statistically different from that detected in subjects receiving placebo.
All volunteers who received the highest dose of the vaccine mounted an excellent immune response, demonstrated by measuring mucosal and systemic responses. This important result proves the vaccine's effectiveness in priming both the mucosal immune system โ" the body's first line of defence โ" as well as the systemic immune system, essential for a successful single dose approach. Injected vaccines elicit a systemic response but a poor mucosal immune response.
Commenting on the result, investigator's Dr Beth Kirkpatrick and Professor Lou Bourgeois said โWe think this oral, single dose freeze-dried vaccine - which was simple to administer โ" is showing great promise providing an excellent basis on which to advance further clinical development.โ
Microscience has used its patented Signature Tagged Mutagenesis (STM) functional genomics technology - which allows the development of safe live attenuated vaccines โ" to develop this new vaccine.
The results of this clinical study further validate the approach of using the oral typhoid vaccine to orally deliver other vaccine antigens for delivery to the immune system.
The company has already developed two further vaccines using this approach. Known as
spi-VEC vaccines, they are both scheduled to enter clinical trials this year. One is a vaccine to protect against a common form of traveller's diarrhoea (ETEC) and the other is a therapeutic vaccine to treat chronic Hepatitis B carriers.
Other conditions that could be prevented, or treated, are also being investigated and include bacterial and viral diseases, allergies and cancer.

Oxford, October 17 2003-- the Council for European National Top Level Domain Registries (CENTR) announced today the creation of the Internet Infrastructure Funding Support Scheme to establish a long lasting financial contribution to the stability of the Internet.
"Stability of the Internet's core function is essential to the CENTR members and the communities they serve" said Paul M. Kane, Chairman of CENTR "Finances are an important element of this stability and we feel the setting up this fund fits perfectly into what CENTR has been doing to support and develop the internet community for the last five years".
Since its creation in March 1998, CENTR has been working with members from 48 countries to contribute to the stability of the Internet in selected areas of importance for the users and the local Internet community of their respective countries as a whole.
- The "Best Practice Guidelines for ccTLD Managers" which have been recently updated to adapt them even closer to the evolution of the needs of users of Internet country code top-level domains (such as .DE, .UK, .FR etc). This Best Practice document has become a trusted reference for Internet Stakeholders.
- Technical Workshops have been organised in essential topics so that exchanges and discussions allow every member to master and benefit from the latest available technology that makes today's Internet and will improve the experience of their local community: Internationalised Domain Names to offer a better flexibility to users who can now access some sites with names written in their own language and script, IPv6 for better and more flexible servers addresses, DNSSEC to enhance security and privacy on the Networkโฆ To improve technical stability of the Internet.
- Legal Workgroups have been created as well to address relevant issues and improve the legal stability of the Internet: Data Privacy, Cyber Squattingโฆ Each of these problems has been of concern to the Internet Community. By sharing them among CENTR members, ccTLDs managers have been able to discover new approaches benefiting the Internet users they representโฆ To improve legal stability of the Internet.
- CENTR also acts as a facilitator between local governments and Internet stakeholders. CENTR regularly meets with Government officials from Europe and the global community and the fact that organisations such as the European Commission are observer members of CENTR, ensures that all the relevant participants can express their views and improve the political stability of the Internet.
The Internet Infrastructure Support Scheme, has to date received contributions amounting to $141,334.50 from .the following ccTLD registries: AC, .AT, .BV, .DE, .GG, .IR, .LU, .NO, .SI, .SJ to ensure the continued stable operation of the IANA function performed by the Internet Corporation for Assigned Names and Numbers (ICANN). Other ccTLDs have indicated their interest in joining the scheme in the coming year.
In addition to this initative CENTR members are providing financial assistance for ccTLD Managers from the African Region to travel to regional meetings and technical workshops. "It is really important that as much as possible that the representatives of the Internet Community worldwide are able to participate in the various meetings and workgroups so we can all work together to continually raise the already exemplary quality of service and security standards of the Internet. That's why we feel it's in CENTR's mission to facilitate outreach to selected countries," concludes Kane.
